Clinically relevant mouse models that represent extremes of the genetic predisposition to anxiety (HAB/NAB/LAB) and comorbid depression-like behavior are used to identify and characterize candidate genes and biomarkers underlying these constructs. Based on comprehensive behavioral and neuroendocrine phenotyping, we focus on molecular-genetic mechanisms contributing to individual variations in trait anxiety up to psychopathology. As exemplified with vasopressin, CRH and glyoxalase-I, genetic polymorphisms and (partially inheritable) epigenetic modifications may interact to causally contribute to the anxious phenotype, thus providing potential targets for diagnosis and individual therapeutic interventions.
